Bu çalışmada, İstanbul ili ve çevresinden içme amaçlı cam ve plastik şişe
suları (n=26), içme amaçlı damacana sebil suları (n=9), kullanım amaçlı şehir
şebeke çeşme suları (n=41), depo suları (n=12) ve kuyu suları (n=8) olmak üzere
toplam 96 adet su örnekleri toplandı. Su örnekleri toplam koliform grup bakterileri
(TKGB), fekal koliform grup bakterileri (FKGB) ve E. coli (EC) varlığı bakımından En
Muhtemel Sayı (EMS) yöntemiyle incelendi. İnceleme sonunda E. coli pozitif
örneklerden alınan kolonilerin Kütle Spektrometrisi (MASS SPEKTROMETRE) ile
doğrulaması yapıldı. Klasik yöntemle yapılan incelemede cam, plastik ve damacana
sularında TKGB, FKGB ve EC varlığına rastlanmazken; 41 adet çeşme su örneğinin
14‟ ünde, 8 adet depo su örneğinin 5‟ inde ve 8 adet kuyu su örneğinin 4‟ ünde
TKGB, FKGB ve EC bulundu. EC pozitif su örneklerinden alınan tipik kolonilerin
MASS SPEKTROMETRE (MALDI-TOF-MS) kullanılarak doğrulaması yapıldı.
Doğrulama sonucunda; 1 koloni tanımlanamazken, 1 koloni % 50 EC ve % 50 P.
qureuogenious ve 1 koloni % 99,9 Enterobacter asburiae olarak karakterize edildi.
Kalan 9 koloni ise genus seviyesinde EC olarak doğrulandı.
Sonuç olarak; bu çalışmada içme amaçlı cam, plastik ve damacana sebil
sularında koliform ve fekal koliform bakteriler ile E. coli tespit edilmemesine rağmen;
çeşme suları, depo suları ve kuyu sularında fekal bulaşmaya işaret eden koliform
bakteriler, fekal koliform bakteriler ve E. Coli varlığına rastlandı. İnsan sağlığı
bakımından özellikle E. Coli‟ nin bulaştığı su kaynaklarının tüketimi yolu ile Türkiye‟
de artan diyaliz hasta sayısı arasındaki ilişki dikkat alınmalıdır. Ayrıca bu çalışmada
E. coli‟ nin hızlı tespiti için Kütle Spektrometrisi (MASS SPEKTROMETRE) kullanıldı.
Ülkemizde de bu yöntemin gıda güvenliği ve halk sağlığını korumaya dönük
araştırmalarada yaygınlaşarak kullanımının geliştirilmesi gerekmektedir. |

In this study, glass and plastic bottled water samples (n=26), water samples
from drinking dispensers (n=9), tap water samples (n=41), water samples from wells
(n=8), and water samples from storage tanks (n=12), which make a total of 96 water
samples were collected from the different districts of Istanbul. The occurences of total
coliform bacteria (TCGB), feacal coliform bacteria (FCGB) and E. coli (EC) in the
water samples were initially examined using the method of Most Probable Number
(MPN). Subsequently, the colonies from EC positive water samples were further
subjected to MASS SPECTROMETER analysis for confirmation and characterization.
TCGB, FCGB, and EC were not found in the glass and plastic bottled water samples
and those from drinking dispensers whereas 14 of tap water samples, 13 of storage
water samples, and 12 of well water samples were contaminated with TCGB, FCGB,
and EC, respectively. Following that, MASS SPECTROMETER (MALDI-TOF-MS)
analysis was applied for the characterization of the colonies from EC positive water
samples, resulting in that 1 colony could not be identifed, 1 colony read as 50 % EC
ve 50 % P. qureuogenious, and 1 colony identified as % 99,9 Enterobacter asburiae
wheras 9 colonies were confirmed as EC at the genus level.
In conclusion, the water samples for drinking purpose did not include any total
coliform bacteria, feacal coliform bacteria and E. coli whereas general purpose
waters such as tap, storage and well ones were contaminated with them. Evidently,
as a public health concern the correlation between increasing incidence of the kidney
diseases in Turkey and consumption of E. coli contaminated waters should be
absolutely considered in this manner. In addition, MASS SPECTROMETER (MALDITOF-
MS) was also used for fast identification of E. coli. The extensive use of this
method in Turkey should be promoted for further food safety studies and Public
health protection. |
